چکیده

objective(s):the mesenchymal stem cells (mscs) have been introduced as appropriate cells for tissue engineering and medical applications. some studies have shown that topography of materials especially physical surface characteristics and particles size could enhance adhesion and proliferation of osteoblasts. in the present research, we studied the distinction effect of 30 and 60 μg/ml of zinc oxide (zno) on differentiation of human mesenchymal stem cells to osteoblast. materials and methods: after the third passage, human bone marrow mesenchymal stem cells were exposed to 30 and 60 μg/ml of zno nanoparticles having a size of 30 nm. the control group has received no zno nanoparticles. on day 15 of incubation for monitoring the cellular differentiation, alizarin red staining and rt-pcr assays were performed to evaluate the level of osteopontin, osteocalsin and alkaline phosphatase genes expression.results:in the group receiving 30 μg/ml of zno nanoparticles, the expression of osteogenic markers such as alkaline phosphatase, osteocalcin and osteopontin genes were significantly higher than both control and the group receiving 60 μg/ml zno nanoparticle. these data also confirmed by alizarin red staining.conclusion:it seems the process of differentiation of mscs affected by zno nanoparticles is dependent on dose as well as on the size of zno.

Effect of hydroalcoholic extract of Cannabis sativa on cell survival and differentiation of mesenchymal stem cells derived from human adipose tissue to osteoblast-like cells

Background and Objective: The resin secretions of Cannabis sativa are called Hashish, which has medicinal and psychological properties. The most important psychoactive compound of this plant is THC (Delta-9-Tetrahydrocannabinol), which can stimulate cannabinoid receptors in the body. This study was carried out to evaluate the effect of hydroalcoholic extract of Cannabis sativa on cell survival ...

In-vitro Differentiation of Human Umbilical Cord Wharton’s Jelly Mesenchymal Stem Cells to Insulin-Producing Cells

Background & Objective: Diabetes is a major chronic metabolic disease in the world. Islet transplantation is a way to treat diabetes. Unfortunately, this method is restricted due to graft rejection and lack of donor islets. Mesenchymal Stem Cells (MSCS) have the ability to differentiate into Insulin-Producing Cells (IPCs). In this study, Human Umbilical Mesenchymal Stem Cells (HUMSCS) were in...

In Vitro Toxic Effects of Zinc Oxide Nanoparticles on Rat Adipose Tissue-Derived Mesenchymal Stem Cells

OBJECTIVE Zinc oxide nanoparticles (ZnO-NPs) are increasingly used in sunscreens, bio- sensors, food additives, pigments, manufacture of rubber products, and electronic materi- als. There are several studies about the effects of NPs on dermal fibroblast or keratino- cytes, but very little attention has been directed towards adipose-derived mesenchymal stem cells (ASCs). A previous study has rev...
